Abstract

Mechanical properties have always been considered one of the primary biomarkers in distinguishing normal cells from tumor cells. The prime purpose of this thesis was to study and analyze the penetration of gold nanoparticles in different stiffness , same sized and varying time-points 3D tumor spheroids using light and electron microscopy techniques. Recently a lot of attention has been given to the culture of cancer spheroids for applications such as drug delivery, nanoparticles interaction, and penetration. Such analysis requires high-quality imaging modalities like transmission electron microscopy (TEM). In this study, a set of spheroids were cryo-sectioned to study the distribution of the particles using light microscopy and another set was analyzed with transmission electron microscopy to study the quantitative distribution of gold nanoparticles and morphological changes in sub-micron features such as cell-cell junctions, nuclear size.
